Stericcorrespondencebetweentheelectrostaticallycomplementarychainsisanimportantfactordeterminingthedefectivenessofthepolyelectrolytecomplexes.Tostudythestericeffectsonpolyelectrolytecomplexbehavior,wedesignedtwoicosapeptide(KKGGandKGKG)withthesamechargedensitybutdifferentchargedistribution.Thecomplexesformedbythetwopeptidewith21ntsingle-strandedoligonucleotides(21ssDNA),21bpdouble-strandedoligonucleotide(21dsDNA),and2000bpsalmontestsDNA(Salmon)inTEbufferwereinvestigatedbylaserlightscatteringandatomicforcemicroscopy.Itwasfoundthatthetwopeptidesfailedtoformcomplexeswith21bpssDNAinthestudiedchargeratio.Intitrationexperiment,whenpeptideswereaddeddropwisetoDNA,boththecomplexformedbyKKGGwith21dsDNAandSalmonshowedstrongerscatteredintensitycomparedwithKGKG.However,when21dsDNAwasaddeddropwisetopeptide,theaggregationformedbyKGKGwasstrongercomparedtoKKGGatthesamechargeratio.AsforSalmon,littledifferencewasobserved.Thekineticsofcomplexbetween21bpDNAandthetwodifferentpeptidesatN/P10bothexperiencedthreestages:aggregation,fluctuationandprecipitation,whileKGKGshowedstrongerassociation.Inconclusion,chargedistributionoftheshortpeptideindeedplayanimportantroleintheircomplexbehavior,especiallywhencomplexwithshortoligonucleotides.
