Silk fi broin (SF), a natural polymer, has fueled wide interest in the biomedical fi eld due to its high biocompatibility, slow biodegradability, nontoxicity, anti-infl ammation, and cell-response and supports cell adhesion and growth. Due to the ample reactive of amino acids, SF could easily be chemically modifi ed via mixture with other active polymers and alter or enhance the physicochemical properties. The ability to be
fabricated into different morphology matrix, namely, particles, fi lms, hydrogels, coatings, etc., also greatly broaden its application for drug delivery systems. Different molecular weights and structures of the drugs could be successfully encapsulated into the SF matrices and achieve sustained delivery/release by adjustments in SF crystallinity, concentration, structure, and design of the delivery systems, which demonstrate a hopeful prospect for the drug delivery system used to treat disease more effi ciently and improve human health. This review will demonstrate different fabrication processes that affect the morphology and properties of SF matrices and discuss a variety of examples for the incorporation of drugs into SF matrices and their drug delivery/release behavior.
